Part 1: Advances in Cancer research using three-dimensional organoid disease models for phenotypic high-throughput therapeutic screening

Advances in cancer research and drug discovery has improved significantly with the application of three-dimensional (3D) organoid disease models for high throughput screening (HTS). The integration of 3D models to determine new treatments has effectively offered a potential bridge between in vitro and in vivo studies to develop new cancer treatments towards personalized medicine. With the usage of 3D ultra-low attachment surfaces, we have successfully screened ex vivo 3D patient-derived primary tumor cancer organoids in a rapid, automated, cost-effective, and highly miniaturized 1536-format against thousands of drugs and/or compounds into a completely scaffold-free 3D environment. This presentation will focus on 3D models from diverse cancers such as pancreas, lung, melanoma, renal carcinoma, rare canine cancer, breast, and glioblastoma and highlight the 3D technologies utilized to perform the drug screening.

Part II: Patient-derived organoids to interrogate phenotypic transitions in metastatic cancer

Metastasis involves non-genetic phenotypic plasticity, leading to therapy resistance and poor clinical outcomes. Using single cell transcriptomics and organoid modeling of normal colon, primary and metastatic colorectal cancer simultaneously resected from the same patients, We define conserved phenotypic transitions and underlying mechanisms, identifying opportunities to target plasticity to treat advanced cancer.
