Corning® Assay Surfaces:
Sulfhydryl-BIND™ (Maleimide) Modified Surface

Corning's Sulfhydryl-BIND™ surface has covalently-linked maleimide groups that covalently couple to sulfhydryl groups (via SH moieties) and are ideal for assays requiring site-directed orientation of a particular biomolecule, especially antibodies (Figure 1). Minimal surface preparation is required: sulfhydryl groups need to be made accessible by the reduction of disulfide bonds or by modification with 2-iminothiolane-HCl (Traut's reagent).

 

Sulfhydryl-BIND Surface Properties 

Applications Assays requiring site-directed orientation of the biomolecule. Useful for the retention or optimization of activity - enzymatic, immunological, etc.

Binding Interaction  Covalent site-specific 
Sample Properties Must have an available sulfhydryl group for covalent coupling via SH moieties
Surface/Sample Preparation  Sulfhydryl groups need to be made accessible by the reduction of disulfide bonds or by modification with 2-Iminothiolane-HCI. Diluent: pH 6-6.5
Blocking Feasibilty & Method    Difficult. Block with 2% BSA in sodium phosphate buffer. Add 10% FBS to antibody diluent.
Performance Criteria   Must demonstrate specific binding: preference for SH, modified goat IgG-Fc over non-modified goat IgG-Fc
