Corning® Assay Surfaces:
High Binding Polystyrene Surface

  High Binding Surface
Figure 1. High binding polystyrene has both hydrophobic and hydrophilic characteristics.

Polystyrene is easily modified to a high binding state by the use of radiation. The radiation effectively incorporates carboxylic acid on the accessible carbons of the "broken" benzene ring. The resulting surface is primarily hydrophobic with intermittent carboxyl groups capable of ionic interactions with positively charged groups on biomolecules (Figure 1). The immobilization mechanism is passive adsorption through hydrophobic and ionic interactions. This is considered a general purpose surface capable of binding medium (>10kDa) and large biomolecules that possess ionic groups and/or hydrophobic regions. Binding capacity is increased as compared to the Medium Binding surface to approximately 400-500ng IgG/cm2 because ionic interactions require that a smaller portion of the molecule be in contact with the surface to obtain stable immobilization.

High Binding Surface Properties

Applications EIA and RIA
# Reactive Sites Binding capacity: 500 ng lgG/cm2 (*See note below.) 1cm2=100 µL volume
Binding Interaction  Hydrophobic and ionic
Sample Properties Medium to high molecular weight. Positively charged. +/- hydrophobic regions
Surface/Sample Preparation None
Blocking Feasibilty & Method    Intermediate. Block with 1% BSA and/or 0.02% Tween 20
Performance Criteria  Well to well CV< 3%. Average high and low wells from the mean +/- 8%
* Note: 500 ng lgG/cm2 is equivalent to monolayer coverage, which should not be exceeded if reproducible results are desired. An observed increase in covalent binding capacity as compared to passive adsorption is due to optimal orientation of the molecule on the surface leading to increased reactivity.
